Protective effect of penehyclidine hydrochloride combined with dexamethasone on acute lung injury induced by endotoxin in Rats

  • 摘要: 目的 探讨盐酸戊乙奎醚(PHC)联合地塞米松(DXM)对大鼠急性肺损伤(ALI)的保护作用.方法 40只SD大鼠随机分为对照组、内毒素(LPS)组、PHC组、DXM组、PHC+DXM组,每组8只.采用尾静脉注射内毒素制备大鼠内毒素性ALI模型后, PHC组和DXM组分别给予PHC和DXM进行干预, PHC+DXM组同时给予PHC和DXM进行干预.各组大鼠于上述处理后6 h留取标本,测定各组肺湿/干值(W/D值)、支气管肺泡灌洗液(BALF)蛋白浓度、中性粒细胞(PMN)计数以及TNF-α和IL-1β的含量,计算肺通透指数,检测肺匀浆髓过氧化物酶(MPO)的活性和丙二醛(MDA)含量.结果 LPS组动脉血气指标PH、PaO2、PaCO2均较Ctrl组明显降低(P<0.01或0.05), 经DXM、PHC或DXM+PHC干预后各指标不同程度升高, DXM+PHC组的PH值、PaO2均不同程度高于DXM组和PHC组,而明显高于LPS组(P<0.01).LPS组肺W/D比、肺通透指数和BALF中PMN计数、MPO、MDA、TNF-α和IL-1β与Ctrl组比较差异有统计学意义(P<0.01), 经DXM、PHC或DXM+PHC干预后各项指标明显改善,与LPS组比较差异有统计学意义(P<0.01或0.05), 而DXM+PHC组的各项指又明显好于DXM组或PHC组(P<0.01或0.05).结论 PHC联合DXM对大鼠LPS性ALI具有明显的保护作用,其效果优于单用PHC或DXM.
