Actin fiber-associated protein 1-antisense RNA 1: a novel oncogenic long noncoding RNA

  • 摘要: 肌动蛋白纤维相关蛋白1-反义RNA 1(AFAP1-AS1)为一个新发现的促癌长链非编码RNA(lncRNA)。AFAP1-AS1定位于人类4号染色体4p16.1区域,被证实在食管癌、胰腺癌、鼻咽癌、肺癌、卵巢癌、结直肠癌、胆道癌、胃癌、肝细胞癌、乳腺癌、舌鳞状细胞癌、视网膜母细胞瘤、胶质瘤和骨肉瘤等多种肿瘤组织中表达上调。AFAP1-AS1过表达与肿瘤大小、淋巴转移、远处转移、TNM分期及不良预后等恶性肿瘤临床病理特征密切相关。此外, AFAP1-AS1可通过调控AFAP1和EMT相关基因、Rho/Rac代谢通路、PTEN/p-AKT通路等促进肿瘤细胞增殖、迁移和侵袭,影响细胞周期进程及抑制细胞凋亡等。
    Abstract: Actin fiber-associated protein 1-antisense RNA 1(AFAP1-AS1)is a newly discovered cancer-promoting long non-coding RNA(lncRNA). AFAP1-AS1 is mapped to the 4p16.1 region of chromosome 4 and found to be upregulated in many malignancies, including esophageal cancer, pancreatic carcinoma, nasopharyngeal carcinoma, lung cancer, ovarian cancer, colorectal cancer, biliary tract cancer, gastric cancer and hepatocellular carcinoma, breast cancer, tongue squamous cell carcinoma, retinoblastoma, glioma and osteosarcoma. Overexpression of AFAP1-AS1 was shown to be associated with several clinicopathological features such as tumor size, lymphatic metastasis, distant metastasis, TNM stage and poor prognosis. Additionally, AFAP1-AS1 may promote cell proliferation, migration and invasion, regulate cell cycle progress and inhibit apoptosis through modulating the expression of AFAP1, EMT related genes, metabolic pathway of Rho/Rac and PTEN/p-AKT pathway.
