Role of epigallocatechin gallate in protecting myocardium of rats with viral myocarditis

摘要:目的 探讨表没食子儿茶素没食子酸酯(EGCG)对病毒性心肌炎小鼠的心肌保护作用。方法 将45只雄性BALA/C小鼠随机分为正常组、对照组及EGCG组, 每组15只。对照组、EGCG组小鼠腹腔注射0.1 mL CVB3病毒液。接种成功后, EGCG组予以EGCG 100 mg/(kg·d)灌胃处理,对照组予以等量的生理盐水灌胃。正常组小鼠腹腔注射等量的生理盐水。饲养1周后,摘除眼球取血检测肌酸激酶同工酶(CK-MB)、心肌肌钙蛋白T(cTnT)的活性,然后处死动物取出心脏, HE染色观察心肌细胞形态学改变。结果 EGCG组小鼠的生存率显著高于对照组; 对照组血清CK-MB、cTnT的活性及心肌病理评分显著高于EGCG组。结论 EGCG对病毒性心肌炎具有一定的保护作用。

Abstract:Objective To investigate the role of epigallocatechin gallate (EGCG) in protecting myocardium of rats with viral myocarditis.Methods A total of 45 male BALA/C rats were randomly divided into normal group, control group and EGCG group, with 15 rats in each group. Rats in the control group and EGCG group were intraperitoneally injected with 0.1 mL CVB3 viral fluid. After successful inoculation, the EGCG group was treated with 100 mg/kg EGCG per day, while the control group was treated with the same amount of saline. Normal rats were injected with the same amount of saline. After feeding for one week, the eyeball blood was removed to detect the activity of creatine kinase isoenzyme (CK-MB) and cardiac troponin T (cTnT). Then the heart was taken out and the morphological changes of cardiac myocytes were observed by HE staining.Results The survival rate of rats in the EGCG group was obviously higher than that in the control group, and the serum CK-MB, cTnT activity and myocardial pathological score in the control group were obviously higher than those in the EGCG group.Conclusion EGCG has a certain protective effect on viral myocarditis.-
