  • 摘要: 铂类作为一种广谱的抗肿瘤药物已得到广泛的认可.奈达铂(NDP)即顺-甘醇酸二氨合铂,是日本研发的第二代铂类抗肿瘤药物,1995年在日本首次获准上市.多年临床研究表明,奈达铂在治疗头颈部癌、肺癌、食管癌、妇科肿瘤等恶性肿瘤中疗效显著,可单药化疗或与多种化疗药物联合并有放疗增敏作用.现就近年奈达铂在恶性肿瘤中的应用进行综述,以供临床参考. 1 奈达铂的作用机制及毒副反应临床研究证实,NDP的抗肿瘤活性与顺铂(DDP)相当[1],抗肿瘤机制与DDP相似,NDP进入细胞后,甘醇酸酯配基上的醇性氧与铂之间的键断裂,水与铂结合,导致离子型物质(活性物质或水合物)的形成,断裂的甘醇酸酯基配基则变得不稳定并被释放,产生多种离子型物质与DNA结合抑制DNA复制,从而发挥抗肿瘤效果.
