Biological effects of recombinant human interferon α on the proliferation of human scar fibroblasts

  • 摘要: 目的 观察重组人α-干扰素(rHu IFN-α)对培养的人瘢痕成纤维细胞增殖的影响,探讨rHu IFN-α对瘢痕成纤维细胞生物学作用.方法 将rHu IFN-α加入瘢痕成纤维细胞培养基内,采用MTT法测定细胞增殖率,用3H-TdR掺入实验检测细胞DNA合成率,应用流式细胞仪分析细胞增殖周期.结果 当rHu IFN-α浓度>4000U/ml时,实验组OD值明显低于对照组(P<0.01).rHu IFN-α组3H-TdR掺入率明显下降,与DMEM组比差异显著(P<0.01).rHu IFN-α组培养24h时瘢痕成纤维细胞进入S期的比例明显降低(P<0.01),至72h时已无显著差别.结论 rHu IFN-α.可以抑制瘢痕成纤维细胞的DNA合成及增殖,其机理为rHu IFN-α.对瘢痕成纤维细胞进入有丝分裂S期的抑制,且其效应呈时间依赖性.
